Creation: An original US route, which in late 1926 ran generally as it does now, plus the US 45E corridor.
Adjustments: About 1927, US 45 was removed from Bethel Purdy Rd; Otis Plunk Rd to McNairy; a bit of McNairy Rd; Although Smith Rd; Clayton Rd to just past Hogwallow Cr and abandoned routing northwest back to today's US 45 just south of TN 199 exists, US 45 may have initially used Litt Wilson Rd to Finger; Main St; Droke Rd/Old Finger Rd to just outside Henderson.
About 1929, US 45 was removed from Circle Hill Dr leaving Selmer. At Fire Tower Rd, the route used to continue due north to the Hopkins/Jackson intersection and TN 5 used Jackson St to Bethel Springs.
In mid-1929, US 45 north was truncated to Threeway, with US 45E continuing into Kentucky. US 45 had not been posted anywhere in Tennessee up to this point, but the old US 45 routing today is still US 45E except: Three Way Lane; Main St through Medina; Highland/College/Main/New St through Milan; Vick Rd south of Bradford and Milan Hwy/Greenfield Hwy through Bradford; Highway Ave through Sharon; US 45E Bus through Martin; TN 43 into South Fulton and Kentucky

About 1930, US 45 was given a direct alignment from Selmer south to the Mississippi line. the original routing was Sticine Rd from the state line area to Caffey; New Hope Rd; Mayflower Rd/Tom Baker Rd/Roy Seay Rd through Gravel Hill; Freddie Davis Rd; Old No 5 S; Ramer-Selmer Rd; Falcon Rd into Selmer.
Also about 1930, US 45 was removed from a bit of Wilson School Rd near the Madison/Chester Line; abandoned routing northwest; Old Henderson Rd to Pinson; a bit of Bear Creek Rd; Simmons Rd plus abandoned routings on both ends; more abandoned routing (some taken over by the modern US 45 SB alignment); both Old Pinson Rds; Harts Bridge Rd
About 1934, US 45 was removed from Main St, White Ave (this is now TN 365), then Garland Rd leaving Henderson, plus Old Jackson Rd and moved to Church Ave north out of Henderson.
About 1958, US 45 was removed from Royal St and Main St in central Jackson and placed on the newly constructed Highland Ave extension south.

About 1980, US 45 was removed from Church Ave through Henderson.
In 1989 (AASHO), US 45E and US 45W's northern convergence was moved south from Fulton KY to the US 51 interchange near South Fulton TN. Thus, US 45 was added back to a piece of roadway within Tennessee, replacing US 45W into Kentucky.

About 1990, US 45 was placed on the existing US 64 bypass of Selmer, then onto the extended new bypass around Bethel Springs. The old route became US 64 Bus through central Selmer; Peach St leaving Selmer; Main St through Bethel Springs.

About 1997, US 45 was removed from what is now the frontage road at the north end of the US 45 Byp near Three Way.
