Here is a look at US 21 history:

About 1928, US 21 was rerouted to cross into Ohio at Marietta, replacing WV 32. The previous routing on the Ohio side became a bit of US 50 (now WV 618) and OH 7.
In 1933, US 21 was given a bypass of Cayce SC, leaving behind SC 2.
In 1935, US 21 was extended south with US 17 to Pocotaligo, then replaced SC 28 to Gardens Corner and Beaufort. US 21 ended at the end of the causeway east of the bridge to Ladys Island.
In 1938, US 21 was given a bypass around central Yemassee SC, leaving behind US 21 ALT (no longer primary routing).
Also about 1938, US 21 was removed from St. Matthews SC by instead replacing the last bit of SC 31, leaving behind SC 6 and SC 26 (now US 601).
In 1948, US 21 was given its bypass of Fort Mill SC, leaving behind US 21 Business.
In 1950, US 21 was given a direct route from Ridgeway to Rock Hill via Great Falls and Fort Lawn, replacing SC 5. Ridgeway to Winnsboro became SC 213 (now SC 34); Winnsboro to Chester became US 321; Chester to Rock Hill became SC 72.
Also in 1950, US 21 was given a direct routing from Orangeburg SC north to Sandy Run, mostly replacing SC 210. The old route became SC 26 (now US 601) from Orangeburg to St Matthews area, then SC 31 (now US 176) to Sandy Run.
In 1953, US 21 was given its bypass of Rock Hill, which was a replacement of US 21 ALT. The route through Rock Hill became US 21 Business (now half secondary, half SC 322).
In 1954, US 21 south was extended from Beaufort to Hunting Island, replacing SC 285. US 21 ended just onto Hunting Island.
About 1956, US 21 was moved onto Super-2 routing around Mooresville, Davidson and Huntersville NC down to the northern edge of Charlotte, leaving behind NC 115.
In 1957, US 21 south was extended to the Hunting Island Lighthouse along the oceanfront.
About 1966, US 21 was given its bypass of Elkin-Jonesville, NC, leaving behind US 21 Business. Most of the bypass was also part of I-77.
Between 1968-70, US 21 south was truncated inland back to Harbor Island.
In 1969, US 21 was rerouted from just inside S.C. to Charlotte, running on today's I-77 from S.C. to Woodlawn Rd, then east to South Blvd. The old route became an extended NC 51 and an extended US 521 (no longer primary routing) into Charlotte.
In 1975, US 21 was added to I-77 in Charlotte from Woodlawn Rd to NC 16 over to Statesville Rd. Woodlawn Rd became part of US 521 while South Blvd north to Morehead became unnumbered; Graham St remaind US 29.
Between 1977-80, US 21 was re-extended back to the original entrance to Hunting Island State Park.
In 1978, US 21 was added to I-77 in Charlotte from NC 16 to La Salle St then over to Statesville Rd. South of La Salle, Statesville became unnumbered.
About 1979, US 21 was truncated to I-81/US 52 in Wytheville, leaving behind US 52 (now VA/WV 598).
Between 1981-86, US 21 south was extended south to the new (current) entrance to Hunting Island State Park. The posting marking the current endpoint has never moved from the previous endpoint.
In 1986 or 1987, US 21 was added to more I-77 in Charlotte from La Salle St to Sunset Rd over to Statesville Rd. This left behind more unnumbered routing.
In 1990, US 21 was given a bypass of Beaufort SC, replacing SC 281 and multiplexing with SC 802. This left behind US 21 Business.
In 2012, US 21's bypass of Beaufort was sent further southwest and around by Parris Island before meeting back up with US 21 at Burton, replacing SC 802 and SC 280. The old route via Ribaut Rd north back to US 21 went back to being SC 281.
Comments: The 1981 official erroneously shows US 21 with US 52 north of Wytheville.
Although it is not designated officially as a scenic byway, I think US 21 is a pretty ride in Virginia.
I believe US 21 is the only US route with three daughters (221, 321, 421) that are longer than the parent.
